生化机理
Colchicine, a tubulin Inhibitor, has been shown to block polymerization of microtubules by binding to tubulin (IC50 = 3.2 μM). Studies indicate that Colchicine also causes apoptosis of cells by acting on tissue plasminogen activators (tPA) and disrupting the cytoskeleton of cells. Exposure of cells to Colchicine has also been reported to result in alterations of Ca2+ concentrations in cells which may also result in apoptosis. Since increased rate of mitosis is associated with cancer cell proliferation, blocking microtubule function with Colchicine has been used as an approach to anti-cancer therapy. For instance, Colchicine blocks the growth of MCF-7 human breast carcinoma cells with an IC50 value of 13 nM. Colchicine produces anti-inflammatory effects by inhibiting neutrophil motility and activity (5 μmol/kg in a mouse model of gouty arthritis) and inhibits the deposition of uric acid, a key aspect in the treatment of gout.
